目的:研究大豆异黄酮对青年雌性大鼠的抗氧化作用并检验其安全性。方法:选用2月龄青年雌性大鼠50只,按体重随机分成5组,每组10只,分别为空白对照组(基础饲料);低(L-SI)、中(M-SI)、高剂量组(H-SI),其大豆异黄酮含量分别为100、200、300 mg/kg bw;雌激素组(EC,己烯雌酚0.5 mg/kg),实验周期为7周。观察各组大鼠实验前、后血清中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、丙二醛(MDA)和谷胱甘肽过氧化物酶(GSH-PX)等活力变化以及子宫雌激素受体(ER-α)含量的变化,并做病理组织学检查,检验其安全性。结果:大豆异黄酮可显著提高各剂量组SOD活力(p<0.05);低、中剂量组CAT活力升高显著,有显著性差异(p<0.05),而在高剂量组没有显著差异;显著降低大鼠MDA含量;对GSH-PX活性的作用不显著。大豆异黄酮可上调大鼠子宫ER-α含量。高剂量组大鼠卵巢发生病理变化。结论:大豆异黄酮可增强青年雌性大鼠的抗氧化能力,其在100~200 mg/kg bw剂量范围对大鼠无不良影响;而当剂量为300mg/kg bw时,对青年雌性大鼠的卵巢产生负面作用。
Objective: To study the antioxidant effect of soybean isoflavones on young female rats and to test their safety. METHODS: Fifty female 2-month-old female rats were randomly divided into 5 groups according to their body weight, 10 in each group. They were blank control group (basal diet); low (L-SI), medium (M-SI), high In the dose group (H-SI), the isoflavone content of the soybean was 100, 200, and 300 mg/kg bw, and that of the estrogen group (EC, diethylstilbestrol 0.5 mg/kg) was 7 weeks. The activities of superoxide dismutase (SOD), catalase (CAT), malondialdehyde (MDA) and glutathione peroxidase (GSH-PX) in the serum of the rats before and after the experiment were observed. As well as changes in uterine estrogen receptor (ER-α) content, and histopathological examinations were performed to check their safety. RESULTS: Soybean isoflavones significantly increased the SOD activity in each dose group (p<0.05). The CAT activity in the low and middle dose groups increased significantly (p<0.05), but there was no significant difference in the high dose group. Reduced MDA content in rats; the effect on GSH-PX activity was not significant. Soy isoflavones can upregulate ER-α content in rat uterus. High dose group rats pathological changes in the ovary. Conclusion: Soybean isoflavones can enhance the antioxidant capacity of young female rats, with no adverse effects on rats in the 100-200 mg/kg bw dose range, and when the dose is 300 mg/kg bw, in young female rats. The ovary has a negative effect.
