在恶性肿瘤的治疗手段中,化疗一直占据着不可替代的地位。联合化疗虽然对一些恶性肿瘤有明显的治疗效果,但对大多数恶性肿瘤效果并不理想,主要原因在于肿瘤细胞对化疗药物产生了多药耐药性。研究证明,MDR的发生是肿瘤细胞从细胞膜、细胞质和细胞核内多种途径,包括细胞信号传导、能量转换、解毒系统、DNA合成等多个环节综合作用的结果,其形成机制非常复杂。MDR的产生机制尚不明确,目前已提出几种可能的机制。根据肿瘤细胞的多药耐药机制,人们先后应用钙离子通道阻滞剂、免疫抑制剂、蛋白激酶抑制剂以及中药等药物作为逆转剂,取得了不同的作用效果。
In the treatment of malignant tumors, chemotherapy has been occupying an irreplaceable position. Although the combination of chemotherapy for some malignant tumors have a significant therapeutic effect, but the effect of most malignant tumors is not ideal, mainly due to the tumor cells on the multidrug resistance of chemotherapy drugs. Studies have shown that the occurrence of MDR is a result of the combined action of tumor cells from multiple aspects of cell membrane, cytoplasm and nucleus, including cell signal transduction, energy conversion, detoxification system and DNA synthesis. The formation mechanism of MDR is very complex. The mechanism of MDR production is not yet clear, and several possible mechanisms have been proposed so far. According to the multi-drug resistance mechanism of tumor cells, people have successively applied different functions such as calcium channel blockers, immunosuppressants, protein kinase inhibitors and traditional Chinese medicines as reversing agents.
