目的探讨细胞外基质(ECM)成分异常与获得性肾囊肿大鼠肾小管间质损害的关系。方法建立化学诱导的获得性肾囊肿疾病(ARCD)大鼠模型。采用免疫组织化学检测不同损害级别的小管间质ECM及α-平滑肌肌动蛋白(SMA)的表达。应用RT-PGR方法检测其mRNA的表达,并分析两者之间的相关性。结果ARCD组不同损害级别的小管间质肾脏组织中,胶原(Col)Ⅰ、Ⅲ、纤连蛋白(FN)及α-SMA表达均显著增加,与小管间质的损害程度呈正相关;层粘连蛋白(LN)的表达组间比较无显著差异,与小管间质的损害程度亦无显著相关性。结论ECM成分异常导致其结构重塑,可能参与了ARCD的形成过程。
Objective To investigate the relationship between extracellular matrix (ECM) component abnormalities and tubulointerstitial damage in rats with acquired renal cysts. Methods A chemically induced rat model of acquired renal cystic disease (ARCD) was established. Immunohistochemistry was used to detect the expression of ECM and α-smooth muscle actin (SMA) in different damage levels. RT-PGR method was used to detect the mRNA expression, and to analyze the correlation between the two. Results The expressions of collagen Ⅰ, Ⅲ, fibronectin (FN) and α-SMA were significantly increased in tubulointerstitial kidneys of different damage levels in ARCD group, which were positively correlated with the degree of tubulointerstitial damage. (LN) expression was no significant difference between the groups, and the degree of tubulointerstitial damage nor significant correlation. Conclusion Abnormal ECM leads to structural remodeling and may be involved in the formation of ARCD.
