为了解橙盖鹅膏菌中分离得到的过氧麦角甾醇对人卵巢癌A2780S细胞增殖的影响及其可能的机制,取对数生长期的A2780S细胞,经1.10、2.19、4.38、8.75、17.5、35μmol/d L等浓度下的过氧麦角甾醇作用,以CCK-8法测试24、48和72 h后细胞增殖;经2.19、4.38、8.75μmol/d L浓度的过氧麦角甾醇作用24 h后,倒置显微镜下观察细胞形态,DAPI染色法观察细胞核形态,Annexin V/PI双染流式细胞术测定细胞的早期凋亡率,PI单染流式细胞术测定细胞周期.结果显示,过氧麦角甾醇作用于人卵巢癌A2780S细胞后,17.5μmol/d L、35μmol/d L浓度组48 h、72 h出现大量细胞死亡,贴壁存活细胞数目明显减少,细胞核呈现核浓缩的高量区域及月牙状的特征,细胞早期凋亡率、细胞S期比例显著增加.本研究表明过氧麦角甾醇对A2780S细胞增殖具有明显抑制作用,可能与其阻滞细胞周期和诱导细胞凋亡有关.
