[目的]探讨黄芩素(Baicalein)与IFN-γ联合诱导HL-60细胞增殖分化的作用及其机制。[方法]实验分为4组:对照组、Baicalein组、IFN-γ组和Baicalein+IFN-γ组。用Baicalein及IFN-γ单独和联合干预HL-60细胞后,采用苔盼蓝拒染法测定细胞增殖抑制率、四氮唑蓝(NBT)还原反应检查细胞的分化以及TUNEL细胞凋亡试剂盒检测细胞凋亡。[结果]与对照组相比,干预组HL-60细胞增殖抑制率、NBT阳性反应率和凋亡指数均明显增高(P﹤0.01),Baicalein+IFN-γ组各项指标均显著高于Baicalein组和IFN-γ组(P﹤0.05)。[结论]Baicalein+IFN-γ具有诱导HL-60细胞分化的协同作用。其作用可能与Baicalein和IFN-γ在诱导某些基因(如ISG)表达时具有协同作用密切相关。
[Objective] To investigate the effect and mechanism of Baicalein and IFN-γ on the proliferation and differentiation of HL-60 cells. [Methods] The experiment was divided into 4 groups: control group, Baicalein group, IFN-γ group and Baicalein + IFN-γ group. After HL-60 cells were treated with Baicalein and IFN-γ alone and in combination, cell proliferation inhibition rate, differentiation of NBT-reduced test cells and apoptosis of TUNEL cells were determined by typan blue exclusion assay. Apoptosis. [Results] Compared with the control group, the proliferation inhibition rate, NBT positive reaction rate and apoptotic index of the intervention group were significantly higher (P<0.01). The Baicalein+IFN-γ group had significantly higher indexes than the Baicalein. Group and IFN-γ group (P<0.05). [Conclusion] Baicalein + IFN-γ had the synergistic effect of inducing differentiation of HL-60 cells. Its effect may be closely related to the synergistic effect of Baicalein and IFN-γ in inducing the expression of certain genes (such as ISG).
