目的 :检测miR-145在非小细胞肺癌(non-small cell lung cancer,NSCLC)中的表达,并探讨其调控神经性钙黏附蛋白(neural cadherin,N-cadherin)在NSCLC侵袭转移中的作用。方法 :实时荧光定量PCR检测31例NSCLC组织中miR-145与Ncadherin的表达水平。采用脂质体法将miR-145 mimics瞬时转染至SPC-A1细胞,RT-PCR和Western blot检测N-cadherin的表达变化;Transwell实验检测转染后SPC-A1细胞的侵袭能力。结果:在NSCLC癌组织中,miR-145与N-cadherin的表达水平分别表现为显著下调和上调,且表达水平与淋巴结转移密切相关。在SPC-A1细胞中,过表达miR-145能降低N-cadherin的表达,并抑制肿瘤细胞的侵袭。结论:miR-145在NSCLC中低表达,其可能通过靶向调控N-cadherin而影响NSCLC的侵袭转移。
Objective: To detect the expression of miR-145 in non-small cell lung cancer (NSCLC) and to explore its role in the regulation of neural cadherin (N-cadherin) in invasion and metastasis of NSCLC. Methods: Real-time fluorescence quantitative PCR was used to detect the expression of miR-145 and Ncadherin in 31 NSCLC tissues. The miR-145 mimics were transiently transfected into SPC-A1 cells by liposome method. The expression of N-cadherin was detected by RT-PCR and Western blot. The invasion ability of SPC-A1 cells was detected by Transwell assay. Results: The expression levels of miR-145 and N-cadherin in NSCLC tissues were significantly down-regulated and up-regulated, respectively. The expression levels of miR-145 and N-cadherin were closely related to lymph node metastasis. In SPC-A1 cells, overexpression of miR-145 decreased N-cadherin expression and inhibited tumor cell invasion. Conclusion: miR-145 is low expressed in NSCLC, which may affect the invasion and metastasis of NSCLC by targeting N-cadherin.
