目的了解5/6肾切除大鼠残余肾组织核转录因子κappaB(NF-κB/IκBα)的表达与肾功能损害的关系,并观察温脾汤对NF-κB/IκBα的影响。方法采用5/6肾切除方法建立肾纤维化大鼠模型,以NF-κB p65亚基单抗及IκBα多抗为一抗,采用二步法免疫组化染色检测大鼠残余肾脏中NF-κB p65/IκBα的表达,结合血清肌酐、尿素氮、24 h蛋白尿等肾功能指标及肾脏病理损害情况,分析它们在肾脏损害中的作用,在此基础上阐明温脾汤肾保护作用与核转录因子-κB/IκB的关系。结果模型组肾组织中NF-κB p65的表达较正常组织显著增高,与尿蛋白及血肌酐增高呈正相关关系,而IκBα表达明显低于正常肾组织。与模型组相比,温脾汤可上调IκBα的表达,抑制NF-κB p65的过度活化。结论温脾汤能减轻5/6肾切除对大鼠肾功能的损伤,这一作用与其能降低肾组织NF-κB表达的异常增强有关。
Objective To investigate the relationship between the expression of nuclear factor-κappaB (NF-κB/IκBα) and renal dysfunction in 5/6 nephrectomized rats and observe the effect of Wenpi Decoction on NF-κB/IκBα. Methods A rat model of renal fibrosis was established by 5/6 nephrectomy. NF-κB p65 subunit monoclonal antibody and IκBα polyclonal antibody were used as primary antibodies. Two-step immunohistochemical staining was used to detect NF-κB in rat residual kidney. The expression of p65/IκBα, combined with renal function indexes such as serum creatinine, urea nitrogen, 24 h proteinuria and renal pathological lesions, analyzed their role in renal damage, and on this basis, elucidated the protective effect and nuclear transcription of Wenpi Decoction. Factor-κB/IκB relationship. Results The expression of NF-κB p65 in kidney tissue of the model group was significantly higher than that of normal tissues, and was positively correlated with the increase of urinary protein and serum creatinine, while the expression of IκBα was significantly lower than that of normal kidney tissue. Compared with the model group, Wenpi Decoction can up-regulate the expression of IκBα and inhibit the over-activation of NF-κB p65. Conclusion Wenpi Decoction can reduce the damage of renal function in rats with 5/6 nephrectomy. This effect is related to the abnormal increase of NF-κB expression in renal tissue.
