用大鼠的再生肝经过匀浆、加热、高速离心和酒精沉淀获得粗制的肝细胞生长因子（RHGF－1），可检测出肝细胞生长因子的活性。RHGF－1再经DEAE离子交换层析，Sephacryls-300凝胶柱层析及FPLCMonoQ色谱，纯化为单体（RHGF－4），经SDS－PAGE证实为单一成份，分子量为18KD。此纯化的RHGF－4相对活性比RHGF－1提高884倍。
Hepatocyte growth factor (HGF) activity was detected by homogenization, heating, high-speed centrifugation and alcohol precipitation of rat’s regenerated liver to obtain crude hepatocyte growth factor (RHGF-1). RHGF-1 was further purified by DEAE ion exchange chromatography, Sephacryls-300 gel column chromatography and FPLCMonoQ chromatography (RHGF-4) and confirmed by SDS-PAGE as a single component with a molecular weight of 18KD. This relative activity of purified RHGF-4 is 884-fold greater than that of RHGF-1.
