目的:以丙烯酸树脂Ⅱ为囊材制备断血流皂苷微球及评价其体外释药情况。方法:用喷雾干燥法制备断血流皂苷微球,以微球的外观形态、收率及包封率为优化指标,用正交试验设计优化制备条件,并对微球进行2种不同介质(人工胃液、人工肠液)中的体外释放度评价。结果:制得的断血流皂苷微球在电镜下,球形表面圆整,粒径分布适宜,收率(97.53±2.39)%,包封率(92.87±4.2)%。断血流皂苷微球在人工胃液中3 h不释药;在人工肠液中1 h内释放不到30%,3 h释药接近60%;此后释药趋于缓慢,8 h释药>80%。结论:所制备断血流皂苷微球有良好的外观形态、较高的收率及包封率。
OBJECTIVE: To prepare saponin microspheres of blood saponin by using acrylic resin Ⅱ as the material of vesicles and evaluate its in vitro drug release. Methods: The blood-saponin microspheres were prepared by spray-drying method. The morphology, yield and entrapment efficiency of the microspheres were optimized. The orthogonal design was used to optimize the preparation conditions. The microspheres were treated with 2 different media Artificial gastric juice, artificial intestinal juice) in vitro release evaluation. Results: The prepared saponin microspheres showed a spherical shape and a good size distribution under the electron microscope. The yield of the saponin microspheres was 97.53 ± 2.39% and the entrapment efficiency was 92.87 ± 4.2%. The blood saponin microspheres did not release drug in artificial gastric juice for 3 h, less than 30% in 1 h artificial artificial intestinal fluid, nearly 60% in 3 h, and then slowly released after 8 h. %. Conclusion: The prepared saponin microspheres have good appearance, high yield and entrapment efficiency.
