目的D-氨基葡萄糖对胶质瘤细胞的生长抑制作用,并探讨其可能的作用机理。方法采用MTT法筛选D-氨基葡萄糖最佳浓度,应用流式细胞仪检测D-氨基葡萄糖对细胞周期的影响。结果D-氨基葡萄糖能明显抑制胶质瘤细胞生长,并且呈现一定的剂量依赖性。3.5mg/LD-氨基葡萄糖表现出最大抑制率,为21.44%。经D-氨基葡萄糖作用后细胞在G2-M期的比例增加,说明细胞停滞在G2-M期,但是未出现凋亡峰。结论D-氨基葡萄糖能抑制人胶质瘤细胞的生长。其作用机理是使细胞生长停滞在G2-M期。
Purpose D-glucosamine inhibits the growth of glioma cells and explores the possible mechanism of action. Methods The optimal concentration of D-glucosamine was screened by MTT method. The effect of D-glucosamine on the cell cycle was detected by flow cytometry. Results D-glucosamine significantly inhibited the growth of glioma cells in a dose-dependent manner. 3.5mg / LD-glucosamine showed the maximum inhibition rate of 21.44%. After D-glucosamine treatment, the proportion of cells increased in G2-M phase, indicating that cells arrested in G2-M phase, but no apoptotic peak appeared. Conclusion D-glucosamine can inhibit the growth of human glioma cells. Its mechanism of action is to arrest cell growth in the G2-M phase.
