目的:研究微小RNA-203表达与骨肉瘤细胞生物学特征的相关性。方法:收集骨肉瘤组织和肉瘤旁正常组织,检测miRNA-203的表达量;培养骨肉瘤细胞株MG63和成骨细胞细胞株hFOB1.19,检测细胞中miRNA-203的表达量;转染miRNA-203的模拟物后6h、12h、18h、24h时,检测细胞增殖、侵袭能力以及相关基因的表达量。结果:骨肉瘤组织中miRNA-203的表达量低于肉瘤旁正常组织,骨肉瘤细胞株MG63中miRNA-203的表达量低于成骨细胞细胞株hFOB1.19;在骨肉瘤细胞株MG63中,miRNA-203的模拟物能够以时间依赖性的方式降低细胞活力、减少侵袭细胞的数目,抑制Bcl-2、cMet、Notch-3、MMP-9的表达。结论:骨肉瘤组织中微小RNA-203的表达量显著降低,增加微小RNA-203的表达能够抑制骨肉瘤细胞的增殖和侵袭,其靶基因为Bcl-2、cMet、Notch-3、MMP-9。
Objective: To study the correlation between microRNA-203 expression and biological characteristics of osteosarcoma cells. Methods: The tissues of osteosarcoma and sarcoma were collected and the expression of miRNA-203 was detected. The osteosarcoma cell line MG63 and osteoblast cell line hFOB1.19 were cultured to detect the expression of miRNA-203 in the cells. At the 6th, 12th, 18th, and 24th hours after the 203 mimic, cell proliferation, invasion ability, and the expression of related genes were detected. Results: The expression of miRNA-203 in osteosarcoma was lower than that in sarcoma. The expression of miRNA-203 in osteosarcoma cell line MG63 was lower than that in osteoblast cell line hFOB1.19. In osteosarcoma cell line MG63, Mimics of miRNA-203 can reduce cell viability, reduce the number of invading cells, and inhibit the expression of Bcl-2, cMet, Notch-3, and MMP-9 in a time-dependent manner. Conclusion: The expression of microRNA-203 in osteosarcoma tissue was significantly decreased. Increasing the expression of microRNA-203 can inhibit the proliferation and invasion of osteosarcoma cells. Its target genes are Bcl-2, cMet, Notch-3, and MMP-9. .
