目的了解新疆地区鼠类中汉坦病毒感染状况及其基因分型。方法于2010年对新疆3个区域(阿拉山口、石河子、乌鲁木齐)捕获鼠类(沙鼠、田鼠、褐家鼠)采集肺脏提取总RNA,经反转录-聚合酶链反应(RTPCR)扩增汉坦病毒M基因片段;将扩增片段克隆和测序分析,确定病毒基因型。结果在312份沙鼠和31份田鼠样本中均未检测到目标基因,在72份褐家鼠样本中,检出阳性样本11份,占15.28%,其基因型均为汉城型。结论首次发现新疆乌鲁木齐市褐家鼠携带汉城型汉坦病毒,应加强该地区肾综合征出血热病例监测和防治工作。
Objective To understand the status and genotyping of Hantaviruses in rodents in Xinjiang region. Methods Total RNA was extracted from the lungs of three rats (Alashankou, Shihezi and Urumqi) collected from the lungs of Xinjiang in 2010 and amplified by reverse transcription - polymerase chain reaction (RTPCR) Hantavirus M gene fragment; the amplified fragment was cloned and sequenced to determine the virus genotype. Results No target gene was detected in 312 gerbils and 31 voles. The positive samples were found in 72 samples of Rattus norvegicus, accounting for 15.28% of the total. The genotypes were all Seoul. Conclusions The brown-backed Hantavirus was found in Urumqi, Xinjiang, for the first time. It is necessary to strengthen the monitoring and prevention of cases of hemorrhagic fever with renal syndrome in this area.
