目的:研究重组人肿瘤坏死因子受体融合蛋白(rhTNFR:Fc)对佐剂性关节炎(adjuvant arthri-tis,AA)大鼠滑膜细胞增殖和分泌功能的影响。方法:完全弗氏佐剂诱导SD大鼠AA模型,3H-TdR掺入法检测滑膜细胞增殖,ELISA法测定滑膜细胞上清中白细胞介素1β(IL-1β),IL-6,IL-17,淋巴细胞毒素(LTα),NF-κB配体的受体或激活因子(RANKL),基质金属蛋白酶-13(MMP-13)水平。结果:rhTNFR:Fc体外给药可以抑制TNF诱导的滑膜细胞增殖,下调IL-1β,IL-6,IL-17,LTα,RANKL,MMP-13的分泌水平。结论:rhTNFR:Fc抑制滑膜细胞增殖和分泌功能可能是其治疗AA的作用机制之一。
Objective: To investigate the effect of recombinant human tumor necrosis factor receptor fusion protein (rhTNFR: Fc) on the proliferation and secretion of synovial cells in adjuvant arthritis-induced arthritis rats. Methods: The rat model of AA was induced by complete Freund’s adjuvant. The proliferation of synovial cells was detected by 3H-TdR incorporation method. The levels of IL-1β, IL-6, IL- -17, lymphocyte toxin (LTα), receptor or activator of NF-κB ligand (RANKL) and matrix metalloproteinase-13 (MMP-13) levels. RESULTS: In vitro administration of rhTNFR: Fc inhibited the TNF-induced synoviocyte proliferation and down-regulated the secretion of IL-1β, IL-6, IL-17, LTα, RANKL and MMP-13. Conclusion: The inhibitory effect of rhTNFR: Fc on the proliferation and secretion of synovial cells may be one of its mechanisms of action in the treatment of AA.
