目的:了解血小板源性生长因子(PDGF)和碱性成纤维细胞生长因子(bFGF)在视网膜增殖膜细胞的表达及其与增殖性玻璃体视网膜病变(PVR)的关系。方法:免疫组化法研究PDGF和bFGF在20例视网膜前膜和1例视网膜下膜(视网膜脱离术后继发)细胞中的表达情况。结果:①15/21(71.4%)例标本对抗PDGF抗体呈阳性反应,14/21(66.7%)例对抗bFGF呈阳性反应;②Eales病和增殖性糖尿病视网膜病引起的血管增殖性前膜(≥61个细胞/视野,1000×)较PVR引起的非血管增殖性前膜(31~60个细胞/视野)内细胞密度高,黄斑前膜(≤30个细胞/视野)最低。结论:PDGF和bFGF在PVR发展过程中起重要作用;PVR膜上增殖细胞可产生bFGF,它是导致严重PVR时玻璃体内bFGF含量异常升高的主要原因。
OBJECTIVE: To investigate the expression of platelet derived growth factor (PDGF) and basic fibroblast growth factor (bFGF) in retinal proliferating cell and its relationship with proliferative vitreoretinopathy (PVR). Methods: The expression of PDGF and bFGF in 20 cases of pre-retinal membrane and 1 case of subretinal membrane (secondary to retinal detachment) were studied by immunohistochemistry. Results: ①15 of 21 (71.4%) specimens showed positive reaction to anti-PDGF antibody and 14 of 21 (66.7%) showed positive response to bFGF. ②Eales disease and proliferative diabetic retinopathy- Membranes (≥61 cells / field, 1000 ×) had higher cell densities than PVR-induced non-vascular proliferative membranes (31-60 cells / field) with the pre-macular membrane (≤30 cells / field) the lowest. CONCLUSION: PDGF and bFGF play an important role in the development of PVR. Proliferating cells in PVR membrane can produce bFGF, which is the main reason for the abnormal increase of bFGF in vitreous in severe PVR.
