目的:建立高效液相色谱法测定紫杉醇微乳的含量。方法:以Agilent TC-C_(18)柱(250 mm×4.6 mm,5μm)为色谱柱,甲醇-乙腈-水(35:35:30)为流动相,流速1.0 ml·min~(-1),检测波长227 nm。结果:紫杉醇在2~60 mg·L~(-1)的浓度范围内线性关系良好(r=0.999 9,n=6),平均回收率为99.80%,RSD=0.77%(n=9)。结论:该法简便、快速、灵敏、准确、专属性强,可用于紫杉醇微乳的质量控制。
Objective: To establish a method for the determination of paclitaxel microemulsion by high performance liquid chromatography. METHODS: The mobile phase consisted of methanol-acetonitrile-water (35:35:30) with a flow rate of 1.0 ml · min -1 on an Agilent TC-C 18 column (250 mm × 4.6 mm, 5 μm) , Detection wavelength of 227 nm. RESULTS: Paclitaxel had a good linearity (r = 0.999 9, n = 6) in the range of 2 to 60 mg · L -1 with an average recovery of 99.80% and RSD of 0.77% (n = 9). Conclusion: The method is simple, rapid, sensitive, accurate and specific and can be used for the quality control of paclitaxel microemulsion.
