目的探讨内皮抑素对人脑胶质肉瘤细胞株GL15中β-catenin和Cyclin D1表达的影响。方法通过质粒转染的方法向GL15细胞中转染pcDNA3.1ES质粒,通过SABC免疫组织化学方法检测转染组与非转染组细胞中β-catenin和Cyclin D1表达。结果空白对照组细胞爬片中β-catenin和CyclinD1的平均光密度值分别为(0.3758±0.0371)和(0.4092±0.0417),而内皮抑素质粒转染组细胞爬片中β-catenin和CyclinD1的平均光密度值分别为(0.2462±0.0275)和(0.2046±0.0357);空白对照组中β-catenin和Cyclin D1的平均光密度值均高于内皮抑素转染组,统计学分析表明在不同组别之间,β-catenin的平均光密度值有显著性差异,其中Cy-clin D1的平均光密度值有极显著性差异。结论内皮抑素在GL15细胞株中可以抑制β-catenin蛋白的过度表达和异位表达,并通过直接和间接的途径,抑制Cyclin D1蛋白的过度表达,从而发挥抑制肿瘤发展的作用。
Objective To investigate the effect of endostatin on the expression of β-catenin and Cyclin D1 in human glioma cell line GL15. Methods The pcDNA3.1 ES plasmid was transfected into GL15 cells by plasmid transfection. The expression of β-catenin and Cyclin D1 in transfected and nontransfected cells was detected by SABC immunohistochemistry. Results The average optical density of β-catenin and CyclinD1 in the cell slide of the blank control group were (0.3758 ± 0.0371) and (0.4092 ± 0.0417), respectively. However, the β-catenin and CyclinD1 (0.2462 ± 0.0275) and (0.2046 ± 0.0357), respectively. The mean optical density of β-catenin and Cyclin D1 in the blank control group were higher than those in the endostatin-transfected group. Statistical analysis showed that in the different groups In addition, the average optical density of β-catenin values were significantly different, which Cy-clin D1 average optical density values were significantly different. Conclusion Endostatin can inhibit the overexpression and ectopic expression of β-catenin protein in GL15 cells and inhibit the overexpression of Cyclin D1 through direct and indirect pathway, which can inhibit tumor growth.
