在LACS缺陷型酵母YB525中对已经克隆的大豆长链脂酰辅酶A合成酶基因(GmLACS)进行表达,并对其功能进行了预测。进化树比对分析显示大豆长链脂酰辅酶A合成酶(GmLACS)与其它植物的LACSs具有高度的同源性;酵母互补测试显示GmLACS蛋白能够互补LACS缺陷型酵母YB525,具有长链脂酰辅酶A合成酶活性;底物偏好性分析显示GmLACS偏好活化大豆中的重要的长链脂肪酸。以上研究结果表明:大豆长链脂酰辅酶A合成酶基因GmLACS编码的蛋白属于LACS家族,能够活化外源脂肪酸并参与了脂肪酸相关的脂类代谢。
The cloned soybean long chain acyl-CoA synthetase gene (GmLACS) was expressed in LACS-deficient yeast YB525 and its function was predicted. Phylogenetic analysis showed that GmLACS was highly homologous to LACSs of other plants. The yeast complementation test showed that GmLACS protein could complement LACS-deficient yeast YB525 with long-chain fatty acyl-coenzyme A synthetase activity; Substrate preference analysis revealed that GmLACS prefers to activate important long chain fatty acids in soybeans. The above results show that the protein encoded by GmLACS of the long-chain fatty acyl-CoA synthetase gene belongs to the LACS family and activates exogenous fatty acids and is involved in fatty acid-related lipid metabolism.
