Schw artz 直接还原法制备99 Tcm Y D P C,分别采用 Lindm o 法与 Scatchard 法测定标记抗体的体外免疫活性分数及亲和常数,经尾静脉注射14.4 M Bq 99 Tcm Y D P C于荷瘤裸鼠,22 h 后进行γ照相及体内放射性分布测定,并用荷结肠癌裸鼠进行对照。研究结果显示:99 Tcm 对 Y D P C的标记率为41% ~65% ,99 Tcm Y D P C的免疫活性分数为 581% ,对人前列腺癌细胞株 P C3 M 的亲和常数为7.71×109 L/m ol。静脉注射99 Tcm Y D P C 于荷人前列腺癌裸鼠22 h 后,除血液与肺外, T/ N T 均大于200,明显高于荷结肠癌裸鼠组。显像结果表明,前列腺癌裸鼠肿瘤部位有明显放射性浓聚,而荷结肠癌裸鼠肿瘤部位无放射性浓聚。表明99 Tcm Y D P C具有良好的体外免疫活性与体内定位性能,放射性核素标记的 Y D P C 有可能应用于前列腺转移癌的临床分期诊断与导向治疗。
Schw artz direct reduction method was used to prepare 99 Tcm Y D P C. The immunological activity and affinity constants of the labeled antibody were determined by Lindm o method and Scatchard method respectively. The tail vein was injected with 14.4 M Bq 99 Tcm Y D P C in tumor-bearing nude mice, 22 h after gamma photography and in vivo radioactivity distribution assay, and the control of nude mice bearing colon cancer. The results showed that 99 Tcm YD P C labeling rate of 41% to 65%, 99 Tcm Y D P C immunosuppressive fraction was 58 1%, on human prostate cancer cell line P C 3 M The affinity constant was 7.71 × 109 L / m ol. The T / N T of 99 Tcm Y D P C was significantly higher than that of nude mice bearing colon cancer after 22 h of 99 Tcm Y D P C injection in nude mice bearing human prostate cancer. The imaging results showed that there was significant radioactive accumulation in the tumor site of the nude mice of prostate cancer, while the tumor site of the nude mice bearing colon cancer had no radioactive accumulation. It is indicated that 99 Tcm Y D P C has good in vitro immunological activity and in vivo localization performance. The radionuclide labeled Y D P C may be used in the clinical staging diagnosis and treatment of prostate cancer.
