目的:观察左归丸对雷公藤多苷(GTW)诱导的生精障碍大鼠模型睾丸Bcl-2、Bax蛋白及m RNA表达的影响,来初步探讨其改善模型大鼠生殖功能的机制。方法:36只SD大鼠随机分为正常组、模型组及左归丸组,每组12只,以GTW 40mg·kg~(-1)·d~(-1)灌胃4周构建SD大鼠为生精障碍模型,左归丸组予以左归丸混悬液6g·kg~(-1)·d~(-1)进行治疗性干预4周。检测各组大鼠的精子计数,并采用免疫组化法、Western blot、RT-PCR检测各组大鼠睾丸组织Bax、Bcl-2蛋白及m RNA的表达,分析比较不同组蛋白表达的差异。结果:左归丸干预治疗后,精子数量明显优于模型组(P<0.05),Bcl-2的蛋白表达显著增多(P<0.05);Bax的蛋白及m RNA表达有下降趋势。结论:左归丸能减轻雷公藤多苷对大鼠生精细胞的损伤程度,上调Bcl-2蛋白的表达,从而抑制生精细胞的凋亡。
Objective: To observe the effects of Zuogui Pill on the expression of Bcl-2, Bax and m RNA in testes induced by tripterygium wilfordii glycosides (GTW) in rats with spermatogenic disorders, and to explore the mechanism of Zuogui Pill in improving the reproductive function of model rats. Methods: Thirty-six SD rats were randomly divided into normal group, model group and Zuogui pill group, with 12 rats in each group. SD rats were treated with GTW 40 mg · kg -1 · d -1 for 4 weeks Zuoguiwan group was given Zuoguiwan suspension 6g · kg -1 (-1) d -1 for therapeutic intervention for 4 weeks. The sperm counts of rats in each group were detected. The expressions of Bax, Bcl-2 and m RNA in testis were detected by immunohistochemistry, Western blot and RT-PCR. The differences among different groups were analyzed. Results: After Zuogui pill intervention, the number of sperm was significantly better than that of the model group (P <0.05), and the protein expression of Bcl-2 was significantly increased (P <0.05). The protein and mRNA expression of Bax decreased. Conclusion: Zuogui pill can reduce the degree of injury of spermatogenic cells in rats, increase the expression of Bcl-2 protein and inhibit the apoptosis of spermatogenic cells.
