目的:探讨microRNA-24(miR-24)对涎腺腺样囊性癌侵袭转移的作用意义。方法 :以唾液腺腺样囊性癌高转移细胞株ACC-M和低转移细胞株ACC-2为实验对象,通过脂质体介导,将miR-24的拟似物(miR-24mimics)转染入研究细胞,逆转录聚合酶链式反应(RT-PCR)检测细胞miR-24的表达,软琼脂克隆形成实验(soft agar assay)观察细胞的增殖能力的改变,Transwell实验检测细胞侵袭能力变化,MTT法检测细胞增殖的改变。结果:高转移细胞株ACC-M转染后的miR-24表达量显著高于低转移细胞株ACC-2,两者相比差异有显著性(P<0.001)。ACC-M细胞转染miR-24 mimics后增殖能力没有影响(P>0.05)。而细胞的侵袭能力降低。结论:低表达miR-24有助于ACC的侵袭能力的发挥,miR-24增高是导致涎腺腺样囊性癌侵袭能力降低的直接原因。
Objective: To investigate the role of microRNA-24 (miR-24) in the invasion and metastasis of salivary adenoid cystic carcinoma. Methods: The murine adenoid cystic carcinoma cell line ACC-M and the cell line ACC-2 with low metastasis were used as experimental subjects. MiR-24 mimics were transfected by liposome The expression of miR-24 was detected by reverse transcriptase-polymerase chain reaction (RT-PCR). The proliferation of cells was observed by soft agar assay. The changes of cell invasion were detected by Transwell assay. MTT assay was used to detect cell proliferation. Results: The expression of miR-24 in ACC-M cells transfected with ACC-M was significantly higher than that in ACC-2 cells transfected with ACC-2. There was significant difference between the two groups (P <0.001). The proliferation of ACC-M cells transfected with miR-24 mimics had no effect (P> 0.05). The cell’s invasive ability is reduced. CONCLUSION: Low expression of miR-24 contributes to the invasion of ACC. The increase of miR-24 is the direct reason for the decrease of invasiveness of salivary adenoid cystic carcinoma.
