目的观察熟地、鳖甲含药血清对大鼠成骨细胞增殖和功能的影响。方法雄性SD大鼠随机分为3组后,分别灌服高浓度熟地、鳖甲煎剂(3.25 g/kg)、低浓度熟地、鳖甲煎剂(1.625 g/kg)及等量容积的蒸馏水,连续灌胃3 d,制得大鼠高、低剂量熟地、鳖甲含药血清及空白对照血清。原代培养并传至第3代经碱性磷酸酶鉴定取得的大鼠成骨细胞,消化计数铺板并分为3组,以上述血清处理72 h,MTT法检测成骨细胞的增殖率,ELISA法检测碱性磷酸酶(ALP)分泌量并以相应OD值进行纠正,并且运用实时荧光定量PCR检测各组成骨细胞核心结合因子α1(Cbfα1)mRNA表达情况。结果低剂量含药血清组成骨细胞增殖率、ALP浓度及Cbfα1 mRNA表达量明显高于其他2组。结论熟地、鳖甲可能是通过提高成骨细胞的增殖潜能而起骨保护作用的。
Objective To observe the effects of Shuda and Biejia serum on the proliferation and function of rat osteoblasts. Methods Male Sprague-Dawley rats were randomly divided into three groups. The rats were drenched with high concentrations of Rehmanniae Decoction (3.25 g / kg), low concentration of Rehmanniae Decoction, turtle shell decoction (1.625 g / kg) and distilled water , Continuous gavage 3 d, prepared rat high and low dose of Rehmannia, turtle containing serum and blank control serum. Primary cultured and passed to the third generation of osteoblasts identified by alkaline phosphatase identified digestion count and planted three groups were treated with the serum for 72 h, MTT assay osteoblast proliferation rate, ELISA Alkaline phosphatase (ALP) secretion was measured and corrected with the corresponding OD value, and the expression of Cbfα1 mRNA in each group was detected by real-time fluorescence quantitative PCR. Results The proliferation rate, the concentration of ALP and the expression of Cbfα1 mRNA in serum of low dose group were significantly higher than those of the other two groups. Conclusion Rehmannia, turtle shell may play a protective role by increasing the proliferation potential of osteoblasts.
