目的 :探讨胃癌发生、发展过程中幽门螺杆菌 (HP)与 p16、Cyclin D1 基因表达的关系。方法 :免疫组化 S- P法检测 p16、Cyclin D1 。结果 :在癌前病变中 ,HP阳性组的 p16阳性表达率低于 HP阴性组 (P<0 .0 1) ,Cyclin D1 阳性表达率高于 HP阴性组 (P<0 .0 1) ;在胃癌中 HP阳性组的 p16阳性表达率与胃癌的浸润、分化程度有关 ,且低于 HP阴性组(P<0 .0 5 )。 Cyclin D1 阳性表达率在 HP阳性和阴性组间无差异 (P>0 .0 5 )。结论 :在胃癌前病变阶段 ,HP可能诱导癌基因 p16失活和原癌基因 Cyclin D1 激活 ,影响了 p16和 Cyclin D1 的表达 ,在胃癌中 ,HP感染对 p16表达缺失仍起着重要作用 ,但不影响 Cyclin D1 的表达。
Objective: To investigate the relationship between Helicobacter pylori (HP) and the expression of p16 and Cyclin D1 genes during the development and progression of gastric cancer. Methods : Immunohistochemical SP method was used to detect p16 and Cyclin D1. RESULTS: In precancerous lesions, the positive expression rate of p16 was lower in the HP positive group than in the HP negative group (P < 0.01), and the positive expression rate of Cyclin D1 was higher than that in the HP negative group (P < 0.01); The positive expression rate of p16 in gastric cancer positive HP-positive group was related to the degree of invasion and differentiation of gastric cancer, and was lower than that of HP-negative group (P<0.05). The positive expression rate of Cyclin D1 was not different between HP positive and negative groups (P>0.05). Conclusion: During precancerous lesions, HP may induce the inactivation of p16 and the activation of protooncogene Cyclin D1, affecting the expression of p16 and Cyclin D1. In gastric cancer, HP infection still plays an important role in the loss of p16 expression. Does not affect the expression of Cyclin D1.
