目的:研究小鼠阳和汤血清增强阿霉素对MCF-7肿瘤细胞抑制作用的机制。方法:用1.08 g.mL-1阳和汤按小鼠体重20 mL.kg-1连续ig 7 d,末次给药1 h采血,分离得含阳和汤1 g.mL-1的血清(YHDS)。用SMY培养细胞24 h,5-溴脱氧尿核苷(BrdU)掺入法检测细胞增殖,软琼脂克隆形成法检测细胞分化,Western blotting检测Bax/Bcl-2表达,原位末端标记检测细胞凋亡,流式细胞术检测细胞线粒体膜电位。结果:小鼠阳和汤血清能促进阿霉素引起的MCF-7细胞生长抑制(抑制率为61.25%±1.66%,P<0.05),上调MCF-7细胞Bax/Bcl-2表达比率,增加细胞凋亡。小鼠阳和汤血清对MCF-7细胞有诱导分化作用(与对照组比较克隆数减少了68.4%,P<0.05)。结论:阳和汤血清有增强阿霉素对MCF-7肿瘤细胞抑制作用,其机制与易化肿瘤细胞凋亡和诱导肿瘤细胞分化有关。
Objective: To study the mechanism of mouse Yanghe decoction enhancing the suppressive effect of doxorubicin on MCF-7 tumor cells. Conclusion: Yanghe Decoction can enhance the suppressive effect of doxorubicin on MCF-7 tumor cells, and its mechanism is related to the facilitation of tumor cell apoptosis and induction of tumor cell differentiation.
