目的:考察黄连、附子及其配伍组对乳鼠心肌细胞的影响,拟从细胞水平阐释黄连、附子有无心肌细胞毒性。方法:采用体外细胞培养,结合MTT比色法研究黄连、附子以及黄连附子1∶1、1∶2配伍组对乳鼠原代心肌细胞的影响。结果:体外实验研究显示:黄连、附子以及黄连与附子配伍各浓度组均能显著抑制心肌细胞OD值(P<0.01),其中单味黄连各浓度组均对心肌细胞具有显著抑制作用,除黄连1组(0.55 mg/mL)外,其余各浓度组对心肌细胞的抑制率均在50%以上,并呈剂量依赖性(P<0.01);等量单味附子组对心肌细胞的抑制效应不如同量单味黄连明显。与等剂量的黄连组比较,配伍各个浓度组则能明显降低等剂量黄连对心肌细胞的抑制率(P<0.01,P<0.05)。结论:黄连减慢心率引起心电紊乱,可能与其导致心肌细胞损伤凋亡而呈现的细胞毒性有关。附子也可引起心肌细胞凋亡,但与黄连配伍后则能缓解黄连单用引起的心电紊乱,改善心率,降低细胞毒性。
OBJECTIVE: To investigate the effects of Coptis chinensis, Aconite and their compatibility group on neonatal rat cardiomyocytes, and to elucidate whether cardiomyocyte cytotoxicity of Coptis chinensis and Aconiti cony at the cellular level. Methods: The effects of Coptis chinensis, Radix Aconiti Lateralis and Coptis chinensis Franch. 1: 1, 1: 2 compatibility group on primary cultured neonatal rat cardiomyocytes were studied by in vitro cell culture and MTT assay. Results: In vitro experiments showed that OD of Coprinus comatus, Rhizoma Coptidis and Rhizoma Coptidis and Coptidis Rhizoma could all significantly inhibit the OD value of cardiomyocytes (P <0.01), and the concentration of Coptidis Rhizoma in each concentration group significantly inhibited cardiomyocytes, 1 group (0.55 mg / mL), the inhibitory rates of cardiomyocytes in other concentrations were above 50% in a dose-dependent manner (P <0.01) As the amount of single-flavored Coptis obvious. Compared with the same dosage of Coptidis group, compatibility of each concentration group could obviously reduce the inhibitory rate of isoproterenol on cardiomyocytes (P <0.01, P <0.05). Conclusion: Coptis slow heart rate caused by electrical disturbance may be related to the cytotoxicity of cardiomyocytes induced by apoptosis. Aconite can also cause myocardial cell apoptosis, but after compatibility with Coptis can relieve monocephalic caused ECG disturbance, improve heart rate and reduce cytotoxicity.
