目的:通过测定鼻息肉中核因子κB亚单位p50(NF-κB-p50)的活性及其与IL-4、γ-IFN表达的关系,来探讨p50对Th1/Th2细胞因子表达调节的可能机制。方法:采用SP免疫组织化学染色方法检测40例鼻息肉患者和20例正常钩突黏膜中IL-4、γ-IFN、p50的表达,以核染阳性率来评估p50活性。对p50活性与IL-4、γ-IFN因子进行直线相关分析。结果:①鼻息肉中p50细胞质、细胞核均有阳性表达,主要位于上皮细胞、炎症细胞及腺上皮细胞细胞质,p50活性显著增强(P<0.01),IL-4表达亦明显增强(P<0.01),γ-IFN表达无明显改变(P>0.05);②Pearson相关性分析,发现p50活性与IL-4表达呈直线正相关关系,r=0.70(P<0.01),而与γ-IFN表达无相关性,r=0.14(P>0.05)。结论:鼻息肉中p50的活性明显增强,p50的激活上调IL-4的表达可能是鼻息肉中Th1/Th2因子失衡的重要环节之一。
Objective: To explore the possible mechanism of p50 on the regulation of Th1 / Th2 cytokines by measuring the activity of nuclear factor κB subunit p50 (NF-κB-p50) and its relationship with the expression of IL-4 and γ-IFN in nasal polyps. Methods: The expressions of IL-4, γ-IFN and p50 in 40 cases of nasal polyps and 20 cases of normal uncinate mucosa were detected by SP immunohistochemical staining. The positive rate of nuclear staining was used to evaluate the activity of p50. Linear correlation analysis between p50 activity and IL-4 and γ-IFN was performed. Results: (1) The p50 cytoplasm and nucleus of nasal polyps were positive, mainly located in the cytoplasm of epithelial cells, inflammatory cells and glandular epithelial cells, the p50 activity was significantly increased (P <0.01), IL-4 expression was also significantly increased (P> 0.05). ②Pearson correlation analysis showed that p50 activity was positively correlated with IL-4 expression, r = 0.70 (P <0.01), but not with the expression of γ-IFN Sex, r = 0.14 (P> 0.05). CONCLUSIONS: The activity of p50 in nasal polyps is significantly increased. The activation of p50 may up-regulate the expression of IL-4, which may be one of the important link of Th1 / Th2 imbalance in nasal polyps.
