目的研究酪氨酸激酶(Syk)的表达与胃癌细胞凋亡的关系。方法RT-PCR检测50例胃癌及癌旁正常胃组织标本中SykmRNA的表达情况,并用流式细胞仪检测胃癌组织肿瘤细胞凋亡,分析Syk表达与胃癌组织肿瘤细胞凋亡的情况。结果所有癌旁胃组织都有Syk基因的表达,而50例胃癌组织中只有11例有表达,癌旁正常胃组织与胃癌组织中Syk基因表达率差异有显著性(P<0·05)。有淋巴结转移的39例胃癌组织中,2例有Syk基因表达,无淋巴结转移的11例胃癌组织中有9例检测到SykmRNA的表达,有淋巴结转移的胃癌组织中SykmRNA的表达率显著降低(P<0·05)。Syk阳性表达的11例胃癌组织的凋亡率为(28·27±4·39)%,显著高于39例Syk阴性的胃癌组织的凋亡率为(3·29±2·37)%(P<0·05)。结论Syk对胃癌的生长和转移可能有抑制作用,诱导胃癌组织肿瘤细胞凋亡可能是其抑癌作用的主要原因之一。
Objective To study the relationship between the expression of tyrosine kinase (Syk) and the apoptosis of gastric cancer cells. Methods The expression of Syk1 mRNA in gastric cancer tissues and adjacent normal gastric tissues was detected by RT-PCR. The apoptosis of gastric cancer cells was detected by flow cytometry. The expression of Syk and the apoptosis of gastric cancer cells were analyzed. Results The expression of Syk gene was detected in all adjacent gastric cancer tissues, while only 11 of 50 gastric cancer tissues were expressed. There was a significant difference in the expression of Syk gene between adjacent normal gastric tissues and gastric cancer tissues (P <0.05). Sym gene expression was found in 2 of 39 gastric cancer tissues with lymph node metastasis. SymphRNA expression was detected in 9 of 11 gastric cancer tissues without lymph node metastasis, and significantly decreased in gastric cancer tissues with lymph node metastasis (P <0 · 05). The apoptosis rate of Syk-negative gastric cancer tissue was (28.27 ± 4.39)%, which was significantly higher than that of Syk-negative gastric cancer tissues (3.29 ± 2.37)% (Syk-negative) P <0 · 05). Conclusion Syk may have an inhibitory effect on the growth and metastasis of gastric cancer. Inducing apoptosis of gastric cancer cells may be one of the main reasons for its anti-tumor effect.
