已发展起来的限制性通道固定相有内表面反相、受保护的疏水相、半透性表面、双区域及混合功能相等多种。由于这些固定相表面具有限制性通道,使得蛋白质这样的大分子不能进入固定相的疏水区而随流动相流出,但药物及其代谢物这样的小分子能进入疏水区而得到分离。这样就能直接血清进样测定药物,免去了脱蛋白及提取等样品预处理环节。本文讨论了这些固定相的特性及其用于血清中药物的测定。
Restricted channel stationary phases have been developed with internal surface inversion, protected hydrophobic phases, semipermeable surfaces, dual zones and multiple mixing functions. Due to the restricted channels on the surface of these stationary phases, macromolecules such as proteins can not enter the hydrophobic region of the stationary phase and flow out of the mobile phase, but small molecules such as drugs and their metabolites can enter the hydrophobic region to be separated. This allows direct injection of serum for the determination of drugs, eliminating the need for deproteinization and sample pretreatment. This article discusses the characteristics of these stationary phases and their use in the determination of serum drugs.
