应用胶原凝胶细胞培养和传代培养的方法培养原位与异位子宫内膜细胞,并经丹那唑处理。在无丹那唑对照培养中,细胞生长良好,形态呈星形,以胞浆突起相互连接。而经两种浓度丹那唑处理后,细胞生长受抑,传代后24h细胞贴壁力受损,部分细胞呈致死性病理改变。丹那唑作用的超微结构变化包括胞浆内大量空泡与脂滴,溶酶体增多,线粒体肿胀与细胞核不规则,核膜内陷。提示胶原凝胶培养系统为较佳的培养子宫内膜细胞的方法,丹那唑可直接作用于人类原位与异位子宫内膜。
Collagen gel cell culture and subculturing methods were used to culture in situ and ectopic endometrial cells and treated with Danazol. In the comparative dansanazole-control cultures, cells grew well, star-shaped, and connected to each other by cytoplasmic processes. After treated with two concentrations of danazol, the cell growth was inhibited, and the adherent ability of the cells was impaired 24h after passage, some of the cells were deadly pathological changes. The ultrastructural changes of the effect of Danazol include a large number of vacuoles and lipid droplets in the cytoplasm, increased lysosomes, mitochondria swelling and irregular nuclei, and nuclear membrane invagination.
