目的:建立兔血浆胞磷胆碱浓度检测的高效液相色谱方法。方法:血浆经高氯酸沉淀,以Venusil MP-C_(18)为色谱柱;流动相为0.1%三氟乙酸,流速为0.8 ml·min~(-1);检测波长282 nm。结果:胞磷胆碱浓度在0.25~40.00 mg·L~(-1)范围内线性关系良好(r=0.999 8);低中高三个浓度(0.50,10.00,30.00 mg·L~(-1))的相对回收率分别为(100.87±3.38)%、(99.53±2.30)%和(100.47±1.47)%;日内RSD分别为3.37%、2.73%和1.76%,日间RSD分别为3.86%、3.34%和1.81%。结论:本方法简便、快速、准确可靠,适用于兔血浆胞磷胆碱浓度的测定及其药物动力学研究。
Objective: To establish a method for the determination of plasma citicoline concentration in rabbit plasma by high performance liquid chromatography. Methods: The plasma was precipitated with perchloric acid, and Venusil MP-C_ (18) was used as the mobile phase. The mobile phase consisted of 0.1% trifluoroacetic acid and the flow rate was 0.8 ml · min ~ (-1). The detection wavelength was 282 nm. Results: The linearity of citicoline in the range of 0.25-40.00 mg · L -1 was good (r = 0.999 8). The three concentrations of low, medium and high concentrations (0.50,10.00,30.00 mg · L -1) ) Were (100.87 ± 3.38)%, (99.53 ± 2.30)% and (100.47 ± 1.47)%, respectively. The intra-day RSD were 3.37%, 2.73% and 1.76% respectively. The daytime RSD were 3.86% and 3.34% % And 1.81%. Conclusion: The method is simple, rapid, accurate and reliable, suitable for the determination of plasma citicoline concentration in rabbits and its pharmacokinetics.
