目的探讨白藜芦醇对黑色素生成的影响及机制。方法以白藜芦醇为受试物,小鼠黑色素瘤细胞B16为研究对象。不同浓度白藜芦醇(12.5、25、50、100、200、400和800μM)处理B16细胞24 h和48 h,CCK8试剂盒检测细胞增殖情况;酶标法测定B16细胞内黑色素含量和酪氨酸酶活性。结果随着白藜芦醇浓度增高,B16细胞存活率降低,各剂量组与对照组之间差异具有统计学意义(P<0.05)。通过probit计算24 h和48 h的细胞半数抑制浓度(IC50)分别为(231.971±0.21)μM和(112.537±0.14)μM。100μM白藜芦醇处理24 h黑色素抑制率为(35.07±4.10)%﹑酪氨酸酶抑制率为(31.63±1.79)%,与对照组相比差异均有统计学意义(P<0.05)。结论白藜芦醇能有效抑制小鼠黑色素瘤细胞B16的增殖,且能够影响细胞内的黑色素的产生,其机制可能与抑制酪氨酸酶的活性有关。
Objective To investigate the effect of resveratrol on melanin production and its mechanism. Methods Resveratrol was used as a test substance and mouse melanoma B16 was selected as the research object. B16 cells were treated with different concentrations of resveratrol (12.5,25,50,100,200,400 and 800μM) for 24 h and 48 h, respectively. The proliferation of B16 cells was detected by CCK8 kit. The levels of melanin and tyramine Acidase activity. Results With the increase of resveratrol concentration, the survival rate of B16 cells decreased. There was a significant difference between each dose group and the control group (P <0.05). The median inhibitory concentration (IC50) at 24 h and 48 h by probit was 231.971 ± 0.21 μM and 112.537 ± 0.14 μM, respectively. The inhibitory rate of melanin treated with 100μM resveratrol for 24 h was (35.07 ± 4.10)% and the tyrosinase inhibition rate was (31.63 ± 1.79)%, which was significantly different from the control group (P <0.05). Conclusion Resveratrol can effectively inhibit the proliferation of B16 melanoma cells, and can affect the intracellular melanin production, the mechanism may be related to the inhibition of tyrosinase activity.
