目的:探讨肺络通合剂对肺纤维化大鼠肺组织病理形态学的影响。方法:选择Wistar大白鼠,气管内注入博莱霉素建立肺纤维化模型。随机分为正常对照组、模型组、中药高、低剂量组和强的松组。除正常对照组外,其余4组均制成肺纤维化动物模型,模型组用生理盐水ig,中药高、低剂量组用肺络通合剂(相当于生药18,9 g.kg-1)ig。强的松组用强的松(2.04mg.kg-1)ig。各组于用药后28,56 d各随机处死一半大鼠取材送检,观察肺泡炎症病理变化,肺纤维化病理变化和纤维连接蛋白(FN)含量。结果:模型组大量胶原纤维增生,呈重度肺纤维化改变,平均病理积分(56 d,下同)为2.50±0.71;肺络通合剂高剂量组呈轻度肺纤维化(1.00±0.82);低剂量组(1.50±0.85)病变程度较中药高剂量组略重,强的松组(1.80±0.92)病变程度较模型组轻,但较中药高剂量组为重;FN含量平均A值(28 d)肺络通高剂量组(49.55±6.57)明显低于模型组(272.11±40.12),且作用优于强的松组(73.25±11.11)。结论:肺络通合剂能有效抑制博莱霉素引起的肺纤维化形成。
Objective: To investigate the effect of “Lungluo Tonghe Mixture” on lung histopathology in rats with pulmonary fibrosis. Methods: Wistar rats were selected and bleomycin was injected intratracheally to establish pulmonary fibrosis model. Randomly divided into normal control group, model group, Chinese medicine high and low dose group and prednisone group. Except normal control group, the other four groups were made animal models of pulmonary fibrosis, the model group with saline ig, Chinese medicine high and low dose group with lungs and collaterals (equivalent to crude drug 18,9 g.kg-1) ig . Prednisone group with prednisone (2.04mg.kg-1) ig. Rats in each group were sacrificed at 28 and 56 days after treatment. The rats were collected and examined for the pathological changes of alveolar inflammation, the pathological changes of pulmonary fibrosis and the content of fibronectin (FN). Results: A large number of collagen fibers hyperplasia were observed in the model group with severe pulmonary fibrosis. The average pathological score (56 days, the same below) was 2.50 ± 0.71 in the model group and mild pulmonary fibrosis (1.00 ± 0.82) in the high - dose group. The lesion degree in low dose group (1.50 ± 0.85) was slightly heavier than that in high dose traditional Chinese medicine group. The lesion degree in prednisone group (1.80 ± 0.92) was lighter than that in model group, but it was heavier than that in high dose traditional Chinese medicine group d) Compared with the model group (272.11 ± 40.12), the high-dose lung-lung group (49.55 ± 6.57) was superior to the prednisone group (73.25 ± 11.11). Conclusion: Feiluotong Mixture can effectively inhibit bleomycin-induced pulmonary fibrosis.
