目的:建立HPLC法测定附子汤中苯甲酰乌头原碱、苯甲酰新乌头原碱、苯甲酰次乌头原碱含量的方法,同时建立10批附子汤的指纹图谱。方法:应用高效液相色谱法,C18色谱柱系统,采用梯度洗脱方法,对附子汤中单酯型生物碱进行了含量测定,同时建立了附子汤高效液相色谱指纹图谱方法。采用中国药典委员会颁布的中药色谱指纹图谱相似度评价软件,对10批附子汤样品的指纹图谱进行相似度评价。结果:在范围内,含量呈良好线性关系。指纹图谱相似度均在0.9以上。结论:建立的HPLC法简便、可靠、准确,可用于附子汤中单酯型生物碱的含量测定和指纹图谱研究,为附子汤相关制剂的质量控制提供参考。
OBJECTIVE: To establish a method for the determination of benzoyl aconitine, benzoyl mesaconitine and benzoyl aconitine in aconite decoction by HPLC. At the same time, the fingerprints of 10 aconite decoctions were established. Methods: The content of monoester alkaloids in aconite soup was determined by HPLC and C18 column system with gradient elution method. At the same time, the HPLC fingerprinting method of aconite soup was established. The fingerprint similarity of 10 samples of aconite soup was evaluated by similarity evaluation software of Chinese traditional medicine chromatography fingerprinting issued by Chinese Pharmacopoeia Commission. Results: Within the range, the content showed a good linear relationship. Fingerprint similarity are above 0.9. CONCLUSION: The established HPLC method is simple, reliable and accurate and can be used for the determination of monoester alkaloids in aconite soup and the study on its fingerprint. It provides a reference for the quality control of aconite-related preparations.
