目的:研究ZD6474对鼻咽癌细胞株CNE2的抑制作用。方法:采用MTT法测定ZD6474的抑制浓度(IC50),流式细胞仪检测ZD6474的凋亡率及细胞周期分布。结果:MTT数据显示,CNE2细胞的抑制率与ZD6474浓度成正相关,随ZD6474浓度升高,抑制作用越明显,IC50=2.531μmol/L;流式细胞术分析显示,ZD6474处理48h使CNE2细胞凋亡率明显升高(24.02±5.90)%,且随浓度升高凋亡率升高越明显,P<0.001。细胞周期结果显示,ZD6474使细胞周期S期(34.96±5.37)%比例下降,G0/G1期(56.25±1.45)%和G2/M期(17.36±5.73)%比例升高。结论:ZD6474能明显抑制CNE2细胞的增殖,促进其凋亡并改变其细胞周期的分布。
Objective: To study the inhibitory effect of ZD6474 on nasopharyngeal carcinoma cell line CNE2. Methods: The inhibitory concentration (ZD6474) of ZD6474 was determined by MTT assay. The apoptosis rate and cell cycle distribution of ZD6474 were detected by flow cytometry. Results: The MTT data showed that the inhibitory rate of CNE2 cells was positively correlated with the concentration of ZD6474. With the increase of ZD6474 concentration, the inhibitory effect was more obvious, IC50 = 2.531μmol / L. Flow cytometry analysis showed that ZD6474 treatment for 48h resulted in apoptosis of CNE2 cells (24.02 ± 5.90)%, and the more the apoptosis rate increased with the increase of concentration, P <0.001. The results of cell cycle showed that ZD6474 decreased the proportion of S phase (34.96 ± 5.37)% in G0 / G1 phase (56.25 ± 1.45%) and G2 / M phase (17.36 ± 5.73)%. Conclusion: ZD6474 can significantly inhibit the proliferation of CNE2 cells, promote its apoptosis and change its cell cycle distribution.
