目的研究干扰素诱导蛋白-10(IP-10)在小年龄儿童(≤5岁)结核病中表达情况,探讨其在儿童结核病中的诊断价值。方法结核病儿童39例和健康儿童25例(均有卡介苗接种史),取外周血5 ml,用淋巴细胞分离液分离外周血单核细胞(PBMC),经ESAT-6刺激,通过实时荧光定量PCR法检测IP-10 m RNA表达水平。结果结核病患儿中IP-10表达较健康儿童明显增高(P<0.005),但肺外结核组患儿IP-10 m RNA表达未见明显增高(P>0.05),IP-10在肺结核组的表达高于肺外结核组,差异无统计学意义(P>0.05)。结论在结核病高发区IP-10检测可能用于诊断有卡介苗接种史的年幼儿童肺结核。
Objective To investigate the expression of interferon-inducible protein-10 (IP-10) in children with tuberculosis under age of 5 years (≤5 years old) and its diagnostic value in childhood tuberculosis. Methods Thirty-nine children with tuberculosis and 25 healthy children with BCG vaccination were enrolled. Peripheral blood was collected from 5 ml. Peripheral blood mononuclear cells (PBMCs) were isolated by lymphocyte separation and stimulated by ESAT-6. Real-time quantitative PCR Method to detect IP-10 m RNA expression levels. Results The expression of IP-10 in children with tuberculosis was significantly higher than that in healthy children (P <0.005), but the expression of IP-10 mRNA was not significantly increased in children with extrapulmonary tuberculosis (P> 0.05) The expression was higher than extrapulmonary tuberculosis group, the difference was not statistically significant (P> 0.05). Conclusion IP-10 test in high incidence of tuberculosis may be used to diagnose tuberculosis in young children with BCG vaccination history.
