目的:探讨c-Met-si RNA体外对喉癌细胞株生长、运动及侵袭的影响。方法:采用阳离子脂质体Lipofectamine2000作为转染试剂,将pSilencer2.0/c-Met-shRNA转染人喉癌细胞株Hep-2,通过RT-PCR及Western-Blot方法检测转染效率,筛选出抑制效果最好的c-Met-siRNA序列;通过MTT法、运动实验及侵袭实验比较转染前后细胞的生长、运动及侵袭能力。结果:pSilencer2.0/c-Met-shRNA转染人喉癌细胞株Hep-2后,c-Met的mRNA和蛋白表达水平显著降低,细胞的生长、运动及侵袭均受到明显抑制。结论:c-Met-siRNA能成功下调靶基因的表达,并能显著抑制喉癌Hep-2细胞的生长、运动及侵袭,是潜在的喉癌治疗新方法。
Objective: To investigate the effects of c-Met-si RNA on the growth, motility and invasion of laryngeal carcinoma cell lines in vitro. Methods: Lipofectamine 2000 was used as a transfection reagent. The pSilencer2.0 / c-Met-shRNA was transfected into human laryngeal carcinoma cell line Hep-2. The transfection efficiency was detected by RT-PCR and Western-Blot. The best c-Met-siRNA sequence was obtained. The growth, motility and invasion ability of cells before and after transfection were compared by MTT assay, exercise assay and invasion assay. Results: After transfection with pSilencer2.0 / c-Met-shRNA, the mRNA and protein expression of c-Met in human laryngeal squamous cell carcinoma cell line Hep-2 was significantly decreased, and the cell growth, motility and invasion were significantly inhibited. CONCLUSION: c-Met-siRNA can successfully down-regulate the expression of target genes and significantly inhibit the growth, movement and invasion of laryngeal cancer Hep-2 cells. It is a potential new therapeutic approach for laryngeal cancer.
