七味白术散对菌群失调腹泻小鼠肠道乳酸杆菌多样性的影响

肠道微生物与人体多项生理功能相关,是中药药效发挥的重要作用者,研究七味白术散对菌群失调腹泻小鼠肠道乳酸杆菌的影响,可以为其疗效机理提供依据.通过灌胃硫酸庆大霉素和头孢拉定对小鼠进行肠道菌群失调造模,然后灌胃给药七味白术散全量传统汤药和50%量超微汤药对造模小鼠进行治疗,提取肠道微生物宏基因组DNA,用乳酸杆菌特异引物对肠道微生物的16S rDNA进行PCR扩增,通过Hha I、Rsa I、Sam I和HindⅢ等酶切后进行ARDRA分析.结果发现:正常组和七味白术散50%剂量超微汤药治疗组OTUs数大于七味白术散传统汤药治疗组,七味白术散作用下乳酸杆菌的多样性指数、均匀度指数及优势度指数分别为0.330、0.091和0.913,均有所恢复;聚类与主成分分析表明,七味白术散对肠道乳酸杆菌具有一定的调整作用,且超微七味白术散50%量汤药的治疗效果优于七味白术散传统汤药.本研究结果说明七味白术散对菌群失调腹泻的疗效与调整肠道乳酸杆菌有关,七味白术散50%超微汤药的效果较之全量传统汤药的效果更好. The diversity index, evenness index and dominance index of lactobacilli were 0.330, 0.091 and 0.913, Analysis of the categories and principal component analysis showed that the effect of Qiweibaizhu San on intestinal Lactobacillus has a certain adjustment effect, and the treatment effect of 50% of the amount of Tangwei Baizhu San decoction is better than that of the traditional Chinese medicine of Qiwei Baizhu San. The results of this study showed that the therapeutic effect of Qiweibaizhu San on bacterial flora of diarrhea was related to the regulation of Lactobacillus in the intestine. The effect of 50% super-decoction of Qiweibaizhu San was better than that of traditional Chinese medicine.
