目的探讨伴有kazal域的富含半胱氨酸的逆转诱导蛋白(RECK)和基质金属蛋白酶-9(MMP-9)在喉鳞癌和癌旁不同距离组织中的表达及其与临床的关系。方法用采用逆转录聚合酶链反应(RT-PCR)法检测38例喉鳞状细胞癌及癌旁不同距离组织中RECK和MMP-9的表达情况,并分析其与临床病理参数的关系。结果 RECK在喉鳞癌组织、癌旁0.2cm、0.5cm及1.0cm的组织中表达强度及表达率依次增高,组间比较有显著统计学差异(P<0.001);MMP-9在喉鳞癌组织、癌旁0.2cm、0.5cm及1.0cm的表达强度及表达率依次降低,组间比较有显著统计学差异(P<0.001)。RECK和MMP-9在癌组织分别与癌旁0.5cm和1.0cm的组织比较及在癌旁0.2cm分别与癌旁0.5cm和1.0cm的组织比较,表达均有统计学差异(P<0.05);但癌组织与癌旁0.2cm的组织中比较及癌旁0.5cm与癌旁1.0cm的组织中比较,表达均无统计学差异(P>0.05)。RECK和MMP-9的表达呈负相关(P<0.05)。喉鳞癌中不同临床分期、病理组织学分级、淋巴转移情况,二者的表达有统计学差异(P<0.05)。结论 RECK在喉鳞癌组织中呈低表达,而MMP-9在喉鳞癌组织中则为高表达;喉鳞癌的安全切缘当以≥0.5cm作为参考标准。
Objective To investigate the expression of RECK and MMP-9 associated with kazal domain in laryngeal squamous cell carcinoma and its adjacent tissues at different distances and its clinical significance . Methods Reverse transcription polymerase chain reaction (RT-PCR) was used to detect the expression of RECK and MMP-9 in 38 cases of laryngeal squamous cell carcinoma and adjacent tissues with different distances, and its relationship with clinicopathological parameters was analyzed. Results The expression intensity and the expression rate of RECK in tissues of laryngeal squamous cell carcinoma, adjacent tissues of 0.2cm, 0.5cm and 1.0cm were significantly higher than that of adjacent tissues (P <0.001). The expression of MMP-9 in laryngeal squamous cell carcinoma The expression intensity and the expression rate of tissues, adjacent tissues of 0.2 cm, 0.5 cm and 1.0 cm decreased in turn, with significant statistical difference (P <0.001). The expression of RECK and MMP-9 in cancer tissue was significantly different from that in the adjacent tissues (0.5cm and 1.0cm) and adjacent tissues (0.2cm and 0.5cm and 1.0cm respectively) (P <0.05) (P> 0.05). However, there was no significant difference between the cancerous tissues and adjacent tissues (0.2cm) and the adjacent tissues (0.5cm to 1.0cm). RECK and MMP-9 expression was negatively correlated (P <0.05). There were significant differences in the clinical stage, pathological grade and lymphatic metastasis between laryngeal squamous cell carcinoma (P <0.05). Conclusions The expression of RECK is low in laryngeal squamous cell carcinoma while the expression of MMP-9 is high in laryngeal squamous cell carcinoma. The safety margin of laryngeal squamous cell carcinoma should be ≥0.5cm as the reference standard.
