γ干扰素抑制兔平滑肌细胞PDGF-β mRNA原位表达和球囊扩张术后再狭窄

目的 阐明干扰素γ (IFN-γ)抑制血管内膜增生和防治经皮冠状动脉成形(PTCA)术后再狭窄的体内机制。方法 建立兔再狭窄血管模型,动态观察体内给予重组干扰素γ (IFN-γ)对损伤后不同时期血管内膜平滑肌细胞(vSMCs)原位表达血小板源性生长因子β mRNA (PDGF-β mRNA)和增殖细胞核抗原(PCNA)的影响。结果 重组IFN-γ显著抑制一周和四周时内膜vSMCs表达PCNA,抑制率分别为88.50%和58.89%;显著抑制新生内膜细胞PDGF-β mRNA的表达,抑制率分别为一周91.45%,二周93.66%和四周的52.92%。结论 球囊损伤后血管局部vSMCs自泌性产生PDGF-β是导致内膜vSMCs增殖和内膜病变形成的主要原因之一。IFN-γ通过下调vSMC原位表达PDGF-β mRNA抑制这一过程,可能用于临床再狭窄的防治。
