【目的】观察金匮肾气丸对庆大霉素(gentamycin,GM)中毒性耳聋豚鼠耳蜗毛细胞钙离子通道蛋白Ca V1.3表达的影响。【方法】将40只健康级白色红目豚鼠随机分为正常组,模型组,金匮肾气丸高、中、低剂量组,模型组豚鼠每日肌肉注射庆大霉素120 mg/kg连续10 d,金匮肾气丸高、中、低剂量组豚鼠分别每日肌肉注射庆大霉素120 mg/kg,同时给予剂量为20.3、13.5、6.08 g·kg-1·d-1的金匮肾气丸水煎剂灌胃给药连续10 d。采用免疫组织化学法检测各组耳蜗毛细胞Ca V1.3阳性表达平均光密度值。【结果】各组豚鼠毛细胞Ca V1.3钙离子通道蛋白平均光密度值比较,模型组较正常组显著降低(P<0.01),中药高剂量组较模型组显著升高,差异有统计学意义(P<0.05)。【结论】金匮肾气丸对GM所致的豚鼠耳中毒具有一定的防治作用,可保护听功能。
【Objective】 To observe the effect of Jinkuishenqiwan on the expression of calcium channel protein Ca V1.3 in gentamycin (GM) -induced deafness guinea pig cochlea hair cells. 【Methods】 Forty healthy white-red guinea pigs were randomly divided into normal group, model group and Jingui Shenqi Wan high, medium and low dose groups. Guinea pigs in model group were intramuscularly injected with gentamicin 120 mg / kg 10 d, Jingui Shenqi pill high, medium and low dose guinea pigs were intramuscular injection of gentamicin 120 mg / kg, while given doses of 20.3,13.5,6.08 g · kg-1 · d-1 of gold Kuishenqi pills decoction administered orally for 10 days. Immunohistochemistry was used to detect the mean optical density of Ca V1.3 expression in cochlear hair cells of each group. 【Results】 Compared with normal group, the average optical density of Ca V1.3 calcium channel protein in hair cells of guinea pigs was significantly lower than that of normal group (P <0.01). The high dose group of traditional Chinese medicine was significantly higher than that of model group Significance (P <0.05). 【Conclusions】 Jinkuishenqi Pills can prevent and cure hearing loss induced by GM in guinea pigs.
