为了研究造血微环境异常在残留白血病发生中的作用和机制 ,采用Dexter型骨髓培养体系形成白血病骨髓基质细胞贴壁层 ,接种HL 6 0细胞共培养 ,用去甲氧基柔红霉素 (IDA)处理 ,观察HL 6 0细胞的活性变化。结果表明 :随着IDA剂量的增加及培养时间的延长 ,HL 6 0细胞活性逐渐减弱 ,与白血病骨髓基质共培养的HL 6 0细胞数明显高于正常对照组 (P <0 .0 5 ) ,骨髓基质细胞层或单纯基质细胞条件培养液体外使IDA对HL 6 0细胞杀伤能力减弱。结论 :白血病骨髓基质有助于HL 6 0细胞抵抗化疗药物 ,对残留白血病的发展具有一定作用。
In order to study the role and mechanism of hematopoietic microenvironment abnormalities in the occurrence of residual leukemia, Dexter-type bone marrow culture system was used to form leukemic bone marrow stromal cells adherent layer. HL60 cells were co-cultured with dexmethoprim daunorubicin (IDA ) To observe the changes of HL 60 cell activity. The results showed that with the increase of IDA dose and the prolongation of culture time, the activity of HL-60 cells decreased gradually. The number of HL-60 cells co-cultured with leukemia bone marrow stroma was significantly higher than that of normal control group (P <0.05) Bone marrow stromal cell layer or simple stromal cell conditioned medium to IDA on HL 60 cells decreased ability to kill. Conclusion: Leukemia bone marrow stromal cells can help HL 60 cells to resist chemotherapeutic drugs and play a role in the development of residual leukemia.
