肿瘤细胞的特点之一是分化异常。二甲基亚砜可诱导某些肿瘤细胞的分化,为肿瘤细胞分化的研究提供了一种方便的途径。 ECa 109细胞株是一种具有恶性上皮细胞特点的食管癌细胞。经固定、包埋将癌细胞超薄切片后进行透射电镜观察。与正常食管上皮细胞相比较,发现了显著的差异。正常食管上皮细胞具有较少的微绒毛、较多的桥粒与张力原纤维。在胞质中不见聚核蛋白体,而为单核蛋白体。核小而圆。而ECa 109食管癌上皮细胞则有较多微绒毛、较少桥粒与张力原纤维。胞质中均为聚核蛋白体。核大而不规则。由于正常细胞与肿瘤细胞的差别在于食管癌细胞是未分化的细胞,我们根据上述观察提出了下列食管上皮细胞分化的指标:1.微绒毛少;2.丰富的张力原纤维;3.较多的桥粒;4.单核蛋白体;5.较规则的细胞核;6.核/质比例小。加入2%DMSO后,ECa 109食管癌细胞出现与正常食管上皮细胞相似的超微结构,并有胞质分化早于胞核的特点。由此表明ECa 109食管癌上皮细胞可被DMSO诱导分化出现正常食管上皮细胞的特点。为应用DMSO等具有分化作用的药物作为治疗药物提供了线索。讨论了可诱导肿瘤细胞分化的药物。 DMSO还可使ECa 109食管癌上皮细胞的高尔基复合器呈环形,外周围之以散在小泡,胞质中有散在大小不等的初级、次级溶酶体。核内易见高电子致密度的带晕颗粒。
One of the hallmarks of tumor cells is abnormal differentiation. Dimethylsulfoxide can induce the differentiation of some tumor cells and provide a convenient way for the study of tumor cell differentiation. After addition of 2% DMSO, ECa 109 esophageal cancer cells appeared similar to the normal esophageal epithelial ultrastructure, and the characteristics of cytoplasmic premature than the nucleus. Thus ECa 109 esophageal epithelial cells can be induced by DMSO induced normal esophageal epithelial cells.
