在pH 6.5的Walpole缓冲溶液中,当以300 nm的光激发时,1,2-萘醌-4-磺酸钠在468 nm处产生1个较强的荧光峰。加入异烟肼后,由于其与1,2-萘醌-4-磺酸钠发生反应,从而使体系颜色加深且荧光信号减弱。在最佳实验条件下,在0.10~8.0μg/mL范围内,体系的荧光信号△F与异烟肼浓度之间有较好的线性关系,检出限为40.2 ng/mL。方法可直接用于测定药物中异烟肼含量,回收率为96.5%~106.3%。
In Walpole buffer solution at pH 6.5, sodium 1,2-naphthoquinone-4-sulfonate produced a strong fluorescence peak at 468 nm when excited with light at 300 nm. After addition of isoniazid, the color of the system is deepened and the fluorescence signal is weakened due to its reaction with sodium 1,2-naphthoquinone-4-sulfonate. Under the optimal experimental conditions, there was a good linear relationship between the fluorescence signal △ F and isoniazid concentration in the range of 0.10 ~ 8.0μg / mL with the detection limit of 40.2ng / mL. The method can be directly used to determine the content of isoniazid in medicine, the recovery rate is 96.5% ~ 106.3%.
