目的研究神经再生抑制性蛋白Nogo在不同发育阶段大鼠中枢神经系统(CNS)中的表达。方法采用RT-PCR方法半定量分析不同发育阶段大鼠(胚胎期14d ̄成年,共7个时间点)脊髓中Nogo mRNA的表达变化。结果Nogo-A mRNA和Nogo-B mRNA在胚胎期14d就可以检测到,在所检查的时间点内,Nogo-A mRNA和Nogo-B mRNA的扩增表现出很恒定的水平,其表达并未有任何特定的时间模式;Nogo-C mRNA的表达比前两者稍晚,在胚胎期21d检测到。结论Nogo在神经系统发育期就已存在,说明Nogo-A分子与髓鞘的成熟化进程关系可能并不很密切,提示Nogo分子在发育阶段具有不同于神经再生抑制作用的其他功能。
Objective To study the expression of Nogo, a neuroregenerative protein, in the central nervous system (CNS) of rats at different developmental stages. Methods Semi-quantitative RT-PCR was used to analyze the changes of Nogo mRNA expression in the spinal cord in different developmental stages (embryonic 14d ~ adult, a total of 7 time points). Results Nogo-A mRNA and Nogo-B mRNA were detected at embryonic day 14, and Nogo-A mRNA and Nogo-B mRNA showed a constant level of amplification at the time points examined, and their expression did not There was no specific time pattern; Nogo-C mRNA was expressed slightly later than the former two and detected at embryonic day 21. Conclusion Nogo existed during the development of the nervous system, indicating that the relationship between Nogo-A and myelin maturation process may not be very close, suggesting that Nogo has other functions that are different from nerve regeneration in developmental stage.
