五子衍宗方对生精障碍模型大鼠附睾精子参数及血清抑制素B的影响

目的:观察五子衍宗方对生精障碍模型大鼠附睾精子参数及血清抑制素B水平的影响。方法:采用随机数字表法将50只雄性SD大鼠分为正常对照组、模型组、甲基睾酮组和五子衍宗方低、高剂量组,每组10只。除正常对照组外,其余各组大鼠按醋酸棉酚50mg/kg体质量灌胃给药,制备生精障碍模型。正常组和模型组大鼠灌服蒸馏水,甲基睾酮组大鼠灌服甲基睾酮混悬液,五子衍宗方低、高剂量组大鼠分别给予低、高浓度(含1.75g、3.5g生药/mL)五子衍宗方混悬液灌胃,每天1次,连续1个月。观察各组大鼠睾丸湿重、睾丸指数及间质细胞数,按WHO标准,采用WLJY-9000荧光染色精子动(静)态图像分析系统检测附睾精子参数,ELISA法检测大鼠血清抑制素B水平。结果:棉酚可造成大鼠睾丸减轻、睾丸指数减小、间质细胞数减少(P<0.01);附睾精子密度降低,活动率和前向运动率(a+b%)下降(P<0.05,P<0.01),血清抑制素B水平降低(P<0.01);五子衍宗方能显著改善大鼠睾丸重量及睾丸指数,增加间质细胞数,改善附睾精子参数(P<0.05,P<0.01),上调血清抑制素B水平(P<0.01),拮抗棉酚造成的大鼠睾丸生精功能损伤。结论:通过上调抑制素B水平,改善睾丸生精功能,可能五子衍宗方治疗不育症的作用机制之一。 Objective: To observe the effect of Wuzi Yanzong decoction on epididymis sperm parameters and serum inhibin B level in rats with spermatogenic dysfunction. Methods: Fifty male Sprague-Dawley rats were randomly divided into normal control group, model group, methyltestosterone group and Wuzi Yanzong Fang low and high dose group, 10 rats in each group. Except the normal control group, rats in other groups were intragastrically administered with ginsenoside 50 mg / kg body weight to prepare spermatogenic disorders model. Rats in normal group and model group were drenched with distilled water and rats in methyltestosterone group were given methyltestosterone suspension. Rats in low and high dose Wuzi Yanzong group were given low and high concentration (including 1.75g, 3.5g crude drug / mL) Wuzi Yanzong side suspension gavage, 1 day, for 1 month. The testis wet weight, testicular index and interstitial cells in each group were observed. The sperm parameters of epididymis were detected by WLJY-9000 fluorescence dynamic spermatozoa (static) image analysis system according to WHO standard. Serum inhibin B Level. Results: Gossypol could reduce testicular weight, decrease testicular index and decrease interstitial cells (P <0.01). The sperm density of epididymal was decreased and the activity and forward movement rate (a + b%) decreased , P <0.01), serum inhibin B level decreased (P <0.01); WZT could significantly improve testicular weight and testicular index, increase interstitial cells and improve sperm parameters of epididymis (P <0.05, P <0.01) ), Up-regulating the serum inhibin B level (P <0.01), antagonizing gossypol-induced rat seminiferous function damage. Conclusion: It is possible that Wu Zi Yan Zong Fang can treat infertility through up-regulating the level of inhibin B and improving the spermatogenic function of testis.
