构建肌肉高表达质粒VR/TPO ,将其在体外定量表达 ,并初步了解其体内表达活性。方法 :以重组TPOcDNA为目的基因 ,构建人VR/TPO高效表达质粒 ,用脂质体法将其转染CHO细胞 ,RT PCR法检测mRNA表达 ,ELISA法检测TPO的表达量 ,并注入体内了解其对巨核系造血的影响。结果 :VR/TPO可在CHO中高效表达 ,其表达量超过 pcDNA3TPO ,并初步发现有促进血小板增加作用。 结论 :VR/TPO为一高效表达TPO质粒 ,在体内外都有一定表达活性。
Construction of muscle high expression plasmid VR / TPO, its quantitative expression in vitro, and preliminary understanding of its in vivo expression activity. Methods: Recombinant TPO cDNA was used as the target gene to construct human VR / TPO high expression plasmids. The recombinant plasmids were transfected into CHO cells by lipofectamine. The expression of TPO mRNA was detected by RT PCR. The expression of TPO was detected by ELISA. The impact of megakaryocytic hematopoiesis. Results: VR / TPO was highly expressed in CHO and its expression was higher than that of pcDNA3TPO. It was found that VR / TPO could promote platelet increase. Conclusion: VR / TPO is an efficient expression plasmid of TPO and has certain expression activity both in vitro and in vivo.
