目的研究当归对β-淀粉样蛋白引起的SH-SY5Y神经细胞中尼古丁受体亚单位在蛋白质水平降低、细胞损伤及脂质过氧化的拮抗作用,了解其神经保护作用的可能机制。方法选取一定浓度的当归预处理SH-SY5Y细胞后,再加入β-淀粉样蛋白处理,用比色法测定细胞MTT还原率,从细胞整体水平了解当归对细胞活性的影响;用Western blotting方法测定神经型尼古丁受体亚单位(α3和α7)蛋白表达水平的变化;用TBA比色法测定脂质过氧化产物(丙二醛)的水平,以了解当归的抗氧化能力。结果安全浓度的当归提取物能减弱β-淀粉样蛋白引起的细胞损伤和脂质过氧化水平升高,对抗β-淀粉样蛋白引起的α3和α7尼古丁受体亚单位蛋白质水平降低。结论当归有一定的神经保护作用。
Objective To investigate the antagonistic effects of Angelica sinensis on protein levels, cell damage and lipid peroxidation in the SH-SY5Y neurons induced by β-amyloid protein, and to understand the possible mechanisms of neuroprotective effect. Methods A certain amount of Angelica sinensis was used to pretreat SH-SY5Y cells, then β-amyloid protein was added. The MTT reduction rate was determined by colorimetry, and the effect of Angelica sinensis on cell activity was determined from the whole cell level; it was determined by Western blotting method. Changes in expression levels of neuronal nicotinic receptor subunit (α3 and α7) proteins; levels of lipid peroxidation products (malondialdehyde) were determined by TBA colorimetry to understand the antioxidant capacity of Angelica sinensis. RESULTS: Safe concentrations of Angelica sinensis extracts attenuated β-amyloid-induced cell damage and increased levels of lipid peroxidation, and reduced β-amyloid-induced protein levels of α3 and α7 nicotinic receptor subunits. Conclusion Angelica has a certain neuroprotective effect.
