为了提高时间相关单光子计数方法测量荧光寿命的准确性,本工作使用SP—70纳秒荧光光谱仪,系统地研究了影响测量准确性的因素.仪器响应函数P(t)主要取决于停止光电倍增管的响应函数H(t)和激发光源的响应函数L(t),响应函数H(t)的畸变主要取决于停止光电倍增管的电压分布,闪光灯上所加激发电压,充气气压,以及灯电极间隙对激发光源的响应函数L(t),有着不可忽略的影响.为此,我们在实验中摸索出了解决办法,给出了仪器最佳实验条件.
In order to improve the accuracy of time-dependent single-photon counting method to measure fluorescence lifetime, this paper systematically studied the factors that affect the accuracy of measurement using SP-70 nanosecond fluorescence spectrometer.The instrument response function P (t) mainly depends on stopping photomultiplication The response function H (t) of the tube and the response function L (t) of the excitation light source, the distortion of the response function H (t) depends mainly on the voltage distribution of the photoplethysmistor to be stopped, the firing voltage applied to the flash, the inflation pressure, Electrode gap on the excitation light source response function L (t), with a non-negligible impact.To this end, we worked out the solution in the experiment, given the best experimental conditions for the instrument.
