目的研究亚砷酸钠(NaAsO2)对人类黑色素瘤G361细胞活性氧自由基(ROS)水平及酪氨酸酶(TYR)活力的影响。方法将体外培养的人类黑色素瘤G361细胞分别暴露于NaAsO2浓度为0.1,0.5及5.0μmol/L2及6h后,测定细胞内ROS水平及TYR活力。结果NaAsO25μmol/L2及6h〔(130.12±3.07)%〕〔(139.55±11.02)%〕染毒组ROS水平明显高于各相应染毒时段对照组水平;染毒2及6h各剂量组TYR活力分别为(99.31±0.81)%,(99.85±1.34)%,(99.52±1.43)%和(99.50±1.88)%,(100.35±1.95)%,(101.68±1.66)%,与各自对照组比较,差异无统计学意义。结论NaAsO2作用下,随着染毒剂量增高,G361细胞内ROS的生成水平增多,呈剂量-效应关系。
Objective To investigate the effect of sodium arsenite (NaAsO2) on reactive oxygen species (ROS) and tyrosinase (TYR) activity in human melanoma G361 cells. Methods The human melanoma G361 cells cultured in vitro were exposed to 0.1, 0.5 and 5.0 μmol / L NaAsO2 for 6 h, and the intracellular ROS levels and TYR activity were measured. Results ROS levels in NaAsO 25μmol / L and 6h 〔(130.12 ± 3.07)%〕 〔((139.55 ± 11.02)%〕 were significantly higher than those in control group at each exposure time; TYR activity at 2 and 6h (99.31 ± 0.81)%, (99.85 ± 1.34)%, (99.52 ± 1.43)% and (99.50 ± 1.88)%, (100.35 ± 1.95)% and (101.68 ± 1.66)%, respectively No statistical significance. Conclusion Under the action of NaAsO2, the generation level of ROS in G361 cells increased with dose increasing, showing dose-effect relationship.
