目的:评价尿酸减轻6-羟基多巴胺(6-OHDA)对PC12细胞的毒性作用。方法:应用PC12细胞制作帕金森细胞模型,分为对照组、尿酸组、6-OHDA组、尿酸+6-OHDA组。采用MTT测定各组PC12细胞活性,免疫荧光法观察各组PC12细胞caspase-3激活情况,流式细胞术检测各组PC12细胞凋亡率。尿酸100~400μmol·L~(-1)不影响PC12细胞生存率,尿酸100~400μmol·L~(-1)可显著提高6-OHDA 50μmol·L~(-1)作用6、12和24 h造成的PC12细胞生存率的下降(P<0.01);尿酸能减少6-OHDA导致的PC12细胞caspase-3激活,降低6-OHDA导致的凋亡率(P<0.05)。结论:尿酸具有减轻6-OHDA对PC12细胞的毒性作用。
OBJECTIVE: To evaluate the effect of uric acid on the cytotoxicity of 6-hydroxydopamine (6-OHDA) on PC12 cells. Methods: PC12 cells were used to make Parkinson’s cell model and divided into control group, uric acid group, 6-OHDA group and uric acid + 6-OHDA group. The activity of PC12 cells in each group was measured by MTT. The activation of caspase-3 in PC12 cells was observed by immunofluorescence. The apoptosis rate of PC12 cells was detected by flow cytometry. Uric acid 100 ~ 400μmol·L -1 did not affect the survival rate of PC12 cells, and uric acid 100 ~ 400μmol·L -1 significantly increased the effect of 50μmol·L -1 6-OHDA for 6, 12 and 24 h (P <0.01). Uric acid decreased the caspase-3 activation induced by 6-OHDA in PC12 cells and decreased the apoptosis rate induced by 6-OHDA (P <0.05). Conclusion: Uric acid can reduce the toxicity of 6-OHDA to PC12 cells.
