目的探讨二苯乙烯苷(TSG)对皮下注射异丙肾上腺素(ISO)引起的C_(57)小鼠心肌肥厚的治疗作用。方法 40只雄性C_(57)小鼠(20~25 g)随机分为4组(每组n=10):Sham组、TSG(120 mg/kg)组、ISO造模组和TSG+ISO组,采用皮下注射ISO制备心肌肥厚模型。给药结束后对各组小鼠进行超声检查检测心脏收缩功能,并测量心脏重量/体重(HW/BW)、肺脏重量/体重(LW/BW)等大体指标。应用HE染色检测各组小鼠心肌平均横截面积的变化。应用PCR技术检测ANP、α-MHC、β-MHC基因表达水平,Western blot检测ANP、α-MHC、β-MHC蛋白表达水平。结果与Sham组比较,ISO组左室射血分数(LVEF)、左室短轴缩短率(LVFS)显著降低,HW/BW、LW/BW明显增大,心肌平均横截面积显著增大,而TSG(120 mg/kg)组的各项指标均无显著变化;与ISO组比较,TSG+ISO组的LVEF、LVFS显著升高,HW/BW、LW/BW明显减小,心肌平均横截面积显著降低;与Sham组比较,ISO组的ANP和α-MHC的表达水平显著升高,β-MHC的表达水平显著降低。与ISO组比较,TSG+ISO组中的ANP和α-MHC的表达水平显著降低,而β-MHC的表达水平显著升高。结论 TSG可以减轻ISO诱导的心肌肥厚,对心脏具有保护作用。
Objective To investigate the therapeutic effect of stilbene glycoside (TSG) on myocardial hypertrophy induced by subcutaneous injection of isoproterenol (ISO) in C57 mice. Methods Forty male C57 mice (20-25 g) were randomly divided into four groups (n = 10 each): Sham group, TSG (120 mg / kg) group, ISO model group and TSG + ISO group , Using subcutaneous injection of ISO preparation of cardiac hypertrophy model. At the end of the administration, the mice in each group were examined with ultrasound to detect the systolic function, and the general indicators such as heart weight / body weight (HW / BW) and lung weight / body weight (LW / BW) HE staining was used to detect changes in the average cross-sectional area of myocardium in each group. Compared with the ISO group, the expression levels of ANP and α-MHC in the TSG + ISO group were significantly decreased, while the expression level of β-MHC was significantly increased. Conclusion TSG can reduce the cardiac hypertrophy induced by ISO, which has a protective effect on the heart.
